Used by many top genomic centers, the Agencourt AMPure system is a highly efficient, easily automated PCR1 purification system that delivers high quality DNA. Requiring no centrifugation or filtration, Agencourt AMPure purifies both small and large PCR extension products in 96- or 384-well formats.
Application: PCR purification
Downstream Application Techniques: Genotyping applications (SNP, fragment analysis), clinical sequencing, sequencing, resequencing
Agencourt AMPure Features:
- Efficient removal of unincorporated dNTPs, primers and salts
- High-quality amplicon purification
- No centrifugation or filtration
- Manual or fully automated formats
- Clean up of a variety of enzymatic reactions including PCR, rolling circle amplification and cDNA labeling

1. PCR reaction 2. Binding of PCR amplicons to magnetic beads 3. Separation of PCR amplicons bound to magnetic beads from contaminants 4. Washing of PCR amplicons with Ethanol 5. Elution of PCR amplicons from the magnetic particles 6. Transfer away from the beads into a new plate
Effective Cleanup:
The Agencourt AMPure system removes unincorporated primers, dNTPs, DNA polymerases and salts used during PCR amplification that can interfere with downstream applications. Agencourt AMPure routinely produces recovery rates greater than 90% for amplicons larger than 200bp and greater than 85% for smaller PCR amplicons, such as the 165bp fragment shown in Figures 1 & 2. Samples purified using Agencourt AMPure are ideal sequencing templates and routinely produce Phred 20 values of greater than 650 bases.

Figure 1: Shown are amplified pGEM2 plasmid fragments of 165, 321, 410, 508 and 967 bp. Half of the reactions were purified using the Agencourt AMPure kit, and the percent recoveries calculated using a PicoGreen2 assay. Samples were run on a 4% agarose gel. Lane MW: 100bp ladder; Lanes 1, 3, 5, 7 and 9: unpurified PCR products; Lanes 2, 4, 6, 8 and 10: Agencourt AMPure-purified samples. |
Figure 2: Fragments sizes of 165, 321, 410, 508 and 967bp from the M13 phage were amplified using PCR, purified using the Agencourt AMPure kit and the percent recovery calculated. |
Scalable and Automation-Compatible
The Agencourt AMPure protocol does not require precipitation, filtration or centrifugation. The Agencourt SPRI technology is easily scaled and automation friendly, allowing both high throughput and format flexibility. Agencourt Bioscience has written and optimized software methods for Beckman Coulter's Biomek® Laboratory Automation Workstations.
