The Agencourt Orapure™ system is a simple, streamlined process for isolating buccal cell genomic DNA (gDNA) for downstream PCR1-based applications, SNP genotyping, sequencing, and re-sequencing. Supporting swab or Scope2 mouthwash collection formats, the SPRI® paramagnetic bead-based technology eliminates the need for filtration or centrifugation. The Agencourt Orapure system effectively and consistently delivers high purity and consistent performance. The process can be run in tube or 96-well formats and is easily automated on either a Beckman FX or NX for multi-microplate processing.

1. Resuspend buccal cells in R1 starting form either a mouthwash or swab sample. 2. Lyse buccal cells in L2.
3. Bind genomic DNA to paramagnetic bead particles using PUR3. 4. Separate beads from contaminants.
5. Wash the magnetic beads with ethanol to remove contaminants. 6. Elute genomic DNA from magnetic particles. 7. Transfer to new plate.

User friendly, Automation Friendly
The Agencourt Orapure system utilizes the power of the Agencourt SPRI® (Solid Phase Reversible Immobilization) paramagnetic bead-based technology that allows for scalability, automation compatibility and a streamlined protocol. There is no need for filtration, centrifugation, or use of chaotropic salts or corrosive detergents. The Agencourt SPRI paramagnetic beads also easily stay suspended in solution and do not catalyze precipitation.

Recovery and Consistent Performance
The Agencourt Orapure process consistently delivers sufficient gNDA to carry out several downstream PCR experiments. Yield can typically exceed 1 µg and recovery testing over on 150-450 µLs of starting volumes using the Scope mouthwash method demonstrated a linear response.

A 2 µL sample of each isolated template from mouthwash was placed into 10 µL PCR reactions with ARF1 primers and Accuprime SuperMix2 and then run on 2% E-Gel12 for 25 minutes.


A 2 µL sample of each isolated template from swab (cytosol brush) was placed into 10 µL PCR reactions with ARF2 primers and Accuprime SuperMix2 and then run on 2% agarose for 25 minutes.

Spectrophotometer absorbance readings were taken from 150, 300 and 450 µL reaction volumes using a NanoDrop2 ND-1000 instrument.
