Amplicon Express' unique library construction methodologies and meticulous quality control procedures produce extremely high quality BAC libraries with average insert sizes greater than 120 Kb (Figure 1) and guaranteed less than 3% error (empty wells + empty clones). Libraries constructed at Amplicon Express can be shipped directly to Agencourt for high throughput BAC end DNA sequencing.

HMW DNA was partially digested with Hind III and ligated into pECBACI. BAC DNA was prepared from random clones, digested with Not I and separated by CHEF gel. More than 90% of the clones contain inserts between 140 Kb and 180 Kb. Marker is Lambda ladder with bands every 50 Kb.

Agencourt End Sequencing
BAC libraries constructed at Amplicon Express are easily integrated into Agencourt's high throughput DNA sequencing pipeline for rapid sequencing. Agencourt has end sequenced more than 750,000 low copy plasmids, including BACs and fosmids. Our pipeline has optimized purification and sequencing protocols for low copy number plasmids producing increased pass rates and Phred20 read lengths.
