Agencourt offers highly flexible genomic shotgun library construction services, including shotgun sequencing of the completed library. We have extensive experience constructing high quality shotgun libraries from a variety of sources and various formats. Library options include:

DNA Sources
  • Mammalian
  • Bacterial
  • Viral
  • Fungal
  • Bacteriophage
  • BAC DNA
Formats
  • 2-5kb insert plasmid libraries in pAGEN High copy vector
  • 10kb insert plasmid libraries in pAGEN Low copy vector
  • 40kb insert fosmid libraries

High Quality Libraries
Our strictly controlled library construction process has several features that ensure we produce high quality libraries.

Construction Process Features:

I. Quality Template DNA
The quality of template DNA is verified by pulse field gel analysis. This ensures that we proceed with high molecular weight starting material, which is necessary to produce high quality libraries.

II. High Cloning Efficiencies
Our hydroshear process produces DNA fragments in a narrow size range, which improves cloning efficiencies and simplifies downstream assembly.

III. Low Background Cloning
BstXI-adapted DNA produces non-complementary ends for highly efficient, low background cloning.

IV. Efficient Cloning of Difficult Regions
Our pAGEN cloning vector with both high and low copy options allows the cloning of small insert and large insert libraries to improve both genome representation and assembly results.

Quality DNA Sequencing
For a complete genome sequencing package, Agencourt offers high quality shotgun DNA sequencing. Once your shotgun library is constructed, it enters our DNA sequencing pipeline. Agencourt plates and picks the appropriate number of clones in order to achieve your desired sequence coverage. The samples are then sequenced on ABI 3730xl DNA sequencers, and data delivered via a secure FTP site. For more information, please see our Whole Genome sequencing service offerings.
